GE Biacore 3000 SPR Biomolecular Interaction Analyzer
The GE Biacore 3000 SPR Biomolecular Interaction Analyzer is an established, label-free system for detailed studies of biomolecular reactions. The Biacore 3000 delivers comprehensive characterization of the interaction, answers questions about the rate constants, affinity, specificity, and determines the active concentration of components. The unique ability to recover and transfer interaction and further characterization makes the system highly applicable to fast identification of unknown interactants. This analyzer is suitable for studying the interactions of proteins, protein conjugates, nucleic acids, lipid micelles, and even larger particles such as viruses and whole cells. Other major applications include measurement of kinetic constants, detection of binding partners and patterns in screening applications, monitoring of enzymatic processes, defining potential drug targets, and more.
The GE Biacore 3000 SPR Biomolecular Interaction Analyzer is designed for real-time biomolecular interaction analysis using surface plasmon resonance (SPR) technology. This technique monitors the formation and dissociation of biomolecular complexes on a sensor surface as the interaction occurs. By covalently attaching one molecule (the ligand) to the surface, the binding of another molecule in a solution (the analyte) with the ligand is followed. Measurements are made under conditions of continuous flow, where the sensor surface forms one wall of the flow cell. The Biacore 3000 measures surface protein concentrations from a few pg/mm^2, and can monitor changes in the surface concentration on a time scale down to 0.1 seconds. This technology allows for determination of binding specificity, kinetics, and concentration with analytes ranging in size from about 150 to 10^6 g/mole (Daltons) or more. Operation of the Biacore 3000 can be preprogrammed for analysis of up to 192 samples ( 2 x 96-well microplates).
