GE AKTA FPLC System
The GE AKTA FPLC System is a
biocompatible, high performance liquid chromatography system for the fast and
easy purification of proteins. Providing flexibility and reliability in every
run, the AKTA system provides a unique combination of flow configuration and
methods support for all chromatographic techniques including gel filtration
(size exclusion) chromatography, anion exchange chromatography, cation exchange
chromatography, hydrophobic interaction chromatography, and affinity
chromatography. The compact and flexible design with an expandable rack allows
the use of longer chromatographic columns and automation kits. The AKTA FPLC
fully meets the demands of modern purification media by utilizing AKTAdesign
and UNICORN to provide reliable results and system versatility. This system features flow rates up to 20 ml/min and pressures up to 5 MPa across one working platform for scale-up production.
The P-920 pump provides highly accurate and reproducible flow rates over the whole pressure range. Its chemical resistance makes it possible to use with corrosive liquids, such as organic solvents, as well as with high salt aqueous solutions. The P-920's high flow range makes it suitable for both analytical and preparative chromatography, and it is designed to work with a wide range of columns and gels from the UPC-900 monitor. The monitor provides precise on-line measurements of UV absorption, pH, and conductivity in liquid chromatography. The UPC-900 offers fixed wavelengths of 214 nm, 254 nm, or 280 nm with fast response. Also included is the M-925 mixer for a dynamic, single chamber unit which provides accuracy and reproducibility. The seven-port motorized valve, INV-907, can easily be used to load or wash the sample loop or wash the pump. The Frac-920 provides fraction collection up to 175 fractions in 12 mm tubes, or 95 fractions using 10-18mm tubes, or 40 fractions with 30 mm diameter tubes, depending on which rack is being used.

- Calibrate pressure and flow rates of both the P-900 pump and P-960/950 pump (if applicable) for precise control
- Replace UV lamp and perform internal inspection tests to confirm complete functionality
- Install a new pH probe and verify the conductivity cell for accurate readings
- Calculate and adjust delay volume in software to reflect system configuration
- Clean or replace the drop sensor, accumulator wash, and height adjuster for consistent collection performance
- Verify fraction collector alignment and calibration with 15mL, 50mL, and microplate receptacles to ensure accuracy
- Run installation tests and thoroughly document all findings for comprehensive record-keeping
